for my 300 i didnt, but a 460 i dont know.

First off, WELCOME to Slick Sixties from South Carolina!
On a '65 or '66, you may need to relocate the transmission support crossmember back a little bit. Mine was less than an inch.
Are you referring instead to the secondary frame cross member that bolts between the radius arm points?
None of this applies if it is going into a '64 or earlier. Nothing is the same between '61-'64 and '65/'66 - except a few pieces of exterior sheet metal, rear axle, some interior trim, and some other odds 'n sods stuff.
Be very particular when looking for interchangeability, or you will get bit. The trucks only look the same on the outside, but underneath there are major differences.

Welcome...
I removed my Tranny cross memeber. Never had a problem. I run a 460 with C6. I think I read somewhere you don't have to remove it but it makes it a PITA... To support the tranny I used a universal tranny mount. Hope this help.
